Morphology:

single, round to polygonal cells in suspension

Medium:

90% RPMI 1640 + 10% h.i. FBS

Subculture:

maintain at 0.3-0.7 x 10⁶cells/ml; split 1:2 to 1:8 every 2 days; keep at 20% FBSfor the first week; seed out at ca. 1 x 10⁶cells/ml in a 6-well-plate or 24-well-plate

Incubation:

at 37 °C with 5% CO₂

Doubling time:

ca. 50-60 hours

Harvest:

maximal density at 1.0 x 10⁶cells/ml
