基础培养基 / Basal Medium:RPMI-1640 Medium(含 2.0 g/L D-Glucose, L-Glutamine)
血清添加 / Serum:10% 优质胎牛血清(FBS)
双抗 / Antibiotics:100 U/mL 青霉素 + 100 µg/mL 链霉素
2. 培养条件与传代规范 / Incubation & Subculture
培养环境 / Conditions:37°C,5% $\text{CO}_2$,饱和湿度。
维持密度 / Density:建议保持 30% – 80% 汇合度,避免细胞长满重叠(到达 90% 以上会导致贴壁变松剥落或降低复苏活力)。
传代操作 / Subculture:
吸除旧培养基,用无钙镁 PBS 温和洗涤 1–2 次。
加入 0.25% Trypsin-0.53 mM EDTA 消化液,37°C 孵育 3–5 分钟,镜检观察细胞脱落变圆。
加入含血清完全培养基终止消化,轻柔吹打均匀。
1000 rpm(200 × g)离心 5 分钟,重悬接种于新培养瓶,传代比例建议 1:2 至 1:4,每 2–3 天传代一次。

1. Background & Context
The HCC1937 cell line (ATCC CRL-2336) was established from a 24-year-old female patient with primary stage IIB grade 3 invasive ductal carcinoma. It represents a widely cited Triple-Negative Breast Cancer (TNBC) and Basal-like cell model. Crucially, HCC1937 is BRCA1-deficient, carrying a germline mutation (5382insC) accompanied by loss of the wild-type allele. It also features a homozygous mutation in TP53 and loss of PTEN expression. Consequently, HCC1937 is a key model for evaluating PARP inhibitors, platinum-based chemotherapy response, and synthetic lethality mechanisms.
