一、 背景与来源说明
ASC52telo 细胞株衍生自正常人脂肪组织提取的间充质干细胞(Adipose-Derived Mesenchymal Stem Cells, ADSCs)。该细胞株通过逆病毒载体介导转染人端粒酶逆转录酶基因(hTERT)构建而成。hTERT 的过表达有效克服了原代间充质干细胞(MSC)在体外长期传代过程中的复制性衰老(Replicative Senescence)问题。ASC52telo 既保留了原代 MSC 的多向分化潜能(成骨、成软骨、成脂分化)与免疫表型特征,又具备无限增殖能力与高度的基因组稳定性,是组织工程、再生医学、干细胞外囊泡(EVs/外体)提取以及干细胞药物开发的标准细胞模型。
二、 细胞生物学特征
组织来源:人(Homo sapiens)脂肪组织(Adipose Tissue)
细胞形态:贴壁生长,呈典型成纤维细胞样(Fibroblast-like)梭形延伸形态
表面标志物表型(MSC Surface Markers):
阳性表达(> 95%):CD29, CD44, CD73 (NT5E), CD90 (Thy-1), CD105 (Endoglin)
阴性表达(< 2%):CD14, CD31 (PECAM-1), CD34, CD45 (LCA), HLA-DR
三系分化潜能(Trilineage Differentiation Potential):
成脂分化(Adipogenesis):在诱导剂作用下形成富含中性脂滴的脂滴细胞(茜素红/油红 O 染色阳性)。
成骨分化(Osteogenesis):形成钙化结节(钙沉积,茜素红 S 染色阳性)。
成软骨分化(Chondrogenesis):在 3D 团聚培养下表达软骨基质(阿利新蓝染色阳性)。
三、 培养条件与传代指南
1. 完全培养基配方
基础培养基:Mesenchymal Stem Cell Basal Medium 或 Alpha-MEM / DMEM/F-12 (1:1)
添加剂与血清:
10% 优质干细胞专用胎牛血清(MSC-Qualified FBS)或 5% 人血小板裂解液(hPL)
2–5 ng/mL 人碱性成纤维细胞生长因子(bFGF / FGF-2)
2 mM L-谷氨酰胺(L-Glutamine)
100 U/mL 青霉素 + 100 ug/mL 链霉素
筛选抗性(用于保持永生化标记):加压维持可选择性添加 G418 或 Hygromycin B(依据构建载体质控说明)。
2. 培养环境与传代规范
培养环境:37°C,5% CO2,饱和湿度。
传代密度控制:细胞密度达到 70% – 80% 时必须传代。切勿让细胞生长至 100% 满融,否则会导致细胞密度抑制并降低后续的分化潜能。
消化操作:使用 0.25% Trypsin - 0.02% EDTA 或 TrypLE Express,37°C 消化 2–3 分钟。加入含血清培养基终止消化,轻柔吹打分散。

1. Background & Context
The ASC52telo cell line is an immortalized human adipose-derived mesenchymal stem cell (ADSC) line established by retroviral transduction of the human telomerase reverse transcriptase (hTERT) gene into primary human ADSCs. Overexpression of hTERT prevents replicative senescence during long-term in vitro passage while maintaining key stem cell attributes. ASC52telo retains multipotent differentiation capacity (adipogenic, osteogenic, and chondrogenic lineages) along with canonical MSC immunophenotypic markers, providing a highly reproducible and scalable non-tumorigenic model for tissue engineering, extracellular vesicle (EV) production, and regenerative medicine research.

3. Culture & Handling Protocols
3.1 Complete Growth Medium Formulations
Basal Medium: Mesenchymal Stem Cell Basal Medium or Alpha-MEM / DMEM/F-12 (1:1).
Supplements & Serum:
10% MSC-Qualified Fetal Bovine Serum (FBS) or 5% Human Platelet Lysate (hPL).
2–5 ng/mL Recombinant Human bFGF (FGF-2).
2 mM L-Glutamine.
Antibiotics: 100 U/mL Penicillin + 100 ug/mL Streptomycin.
3.2 Incubation & Subculture Parameters
Incubation Conditions: 37°C, 5% CO2, humidified atmosphere.
Subculture Confluency: Subculture when cells reach 70% – 80% confluency. Avoid letting cultures reach 100% saturation to prevent contact inhibition and loss of differentiation potential.
Detachment: Treat with 0.25% Trypsin - 0.02% EDTA or TrypLE Express at 37°C for 2–3 minutes. Neutralize with serum-containing medium and gently dissociate into a single-cell suspension.
