Genetic Studies:Threonine Biosynthesis Gene Cluster: The hom, thrB, and thrC genes involved in threonine biosynthesis have been cloned and sequenced, encoding homoserine dehydrogenase, homoserine kinase, and threonine synthase, respectively. These genes are organized in the order hom-thrC-thrB, similar to Streptomyces coelicolor -1-9
ask-asd Operon: The first two genes of the threonine pathway, ask and asd, encoding aspartokinase and aspartate semialdehyde dehydrogenase, have been cloned and sequenced. They are organized as a bicistronic operon. Disruption of asd significantly decreases AVG production, supporting the involvement of this operon in AVG biosynthesis -5-8
AVG Biosynthetic Pathway: Disruption of the hom gene completely blocks AVG biosynthesis, indicating that homoserine is the branching point of the aspartic acid-derived biosynthetic route leading to AVG. The four carbon atoms of the vinylglycine moiety of AVG derive from homoserine -7-10
