• 家族系谱: 该细胞系是 Mac-1 (ACC 990) 的系列姐妹细胞系（Mac-1 早于其2年建立），也是 Mac-2A (ACC 991) 的同步姐妹细胞系。这些姐妹细胞系在核型和免疫表型上存在差异。

• 形态学: 悬浮生长。主要是带有大量“毛刺/尖刺 (spikes)”的圆形细胞，呈单体或小细胞簇生长；可见少量较大细胞。

• 培养基: 85% RPMI 1640 + 15% 热灭活胎牛血清 (h.i. FBS)。

• 传代比例: 按照 1:2 至 1:8 的比例传代，每 2-4 天一次；维持细胞密度在 0.1-0.6 x 10⁶ cells/ml。

• 接种密度: 在 6 孔或 24 孔板中建议起始接种量约为 0.1-0.4 x 10⁶ cells/ml。

• 培养条件: 37 °C, 5% CO₂。

• 倍增时间: 约 24-50 小时（取决于血清浓度和细胞密度）。

• 最大密度: 约 2 x 10⁶ cells/ml。

• 冷冻保存: 70% 培养基, 20% FBS, 10% DMSO。

• Establishment: Established in 1987 from an ulcerating skin lesion of a 47-year-old male with cutaneous large-cell T-cell lymphoma. This patient had been diagnosed with Hodgkin lymphoma 12 years prior.

• Sister Cell Lines: This is a serial sister cell line of Mac-1 (ACC 990) and a simultaneous sister cell line of Mac-2A (ACC 991). While they share the same origin, they differ in karyotypes and immunophenotypes.

• Morphology: Suspension culture. Mainly round cells with numerous "spikes," growing as single cells or small clusters.

• Culture Medium: 85% RPMI 1640 + 15% heat-inactivated FBS.

• Subculture: Split 1:2 to 1:8 every 2-4 days; maintain the culture at a density of approx. 0.1-0.6 x 10⁶ cells/ml.

• Seeding Density: Seed at 0.1-0.4 x 10⁶ cells/ml in 6- to 24-well plates.

• Incubation: 37 °C with 5% CO₂.

• Doubling Time: Approx. 24-50 hours (sensitive to FBS concentration and density).
