Organism: Mouse (Mus musculus), FVB/N breed.
Tissue Source: Ovary, surface epithelium (in situ).
Cell Type: Epithelial.
Growth Mode: Adherent, transitioning from an epithelial "cobblestone" to a mesenchymal morphology during transformation.
Doubling Time: Approximately 13 hours (highly proliferative compared to the parental 48-hour doubling time).
Basal Medium:RPMI-1640 or DMEM.
Supplements: 8–10% Fetal Bovine Serum (FBS), 1% Penicillin/Streptomycin, 1mM Sodium Pyruvate, and 0.1mM Non-Essential Amino Acids (NEAA).
Environment: 37°C in a humidified atmosphere of 5% CO₂.
HGSOC Markers: STOSE-derived tumors are typically positive for pan-cytokeratin (CK), WT1, and PAX8, while remaining negative for inhibin.
Genetic Profile: Characterised by the loss of Cdkn2a and upregulation of Ccnd1 (cyclin D1), mirroring changes in human TCGA ovarian cancer datasets.
Tumorigenicity: Forms tumors and ascites in both SCID and immunocompetent FVB/N mice within approximately 47–48 days following intraperitoneal injection.
STOSE.M1 luc: A luciferase-labeled version used for real-time, quantitative tracking of tumor progression and metastasis via IVIS imaging.
SCA1+ Population: A subpopulation with enhanced tumor-initiating capacity, forming palpable tumors roughly 8 days faster than SCA1- cells.
