BioVector TM AML14.3D10 细胞株是源自人造血髓系白血病的亚克隆,是目前研究嗜酸性粒细胞(Eosinophils)最常用的体外模型。与未分化的亲本 AML14 不同,3D10 亚系已自发分化为嗜酸性中幼粒细胞阶段。

培养指南
  • 培养基:RPMI 1640 + 8% 胎牛血清 (FBS) + 2mM L-谷氨酰胺 + 1mM 丙酮酸钠 + 50μM β-巯基乙醇

  • 接种密度: 维持在 3 x 10⁵ 至 1 x 10⁶ cells/mL 之间。

  • 关键点: 切勿让密度超过 1 x 10⁶ cells/mL。过密会导致细胞脱颗粒,释放出的主碱性蛋白 (MBP) 等毒性物质会引发连锁死亡。

  • 传代比例: 每 3–4 天按 1:2 或 1:3 比例传代。

生物学特征
  • 颗粒蛋白: 高表达主碱性蛋白 (MBP)、嗜酸性粒细胞过氧化物酶 (EPO) 和嗜酸性粒细胞衍生神经毒素 (EDN)。

  • 受体: 表达 GM-CSFIL-5 受体。需注意该亚系已失去 IL-3 受体 α 亚基。

  • 细胞因子依赖: 增殖不依赖外源细胞因子,但对 IL-5 和 GM-CSF 有明显的生物学反应。

处理与保存
  • 冻存液: 90% FBS + 10% DMSO。

  • 复苏注意事项: 初始复苏存活率可能较低(20%–30%)。建议在复苏后的前 72 小时内增加补液频率以稀释死亡细胞释放的颗粒毒素。

The AML14.3D10 cell line is a human hematopoietic myeloid leukemia-derived subclone widely used as a research model for eosinophils. Unlike the parental AML14 line, which is undifferentiated, the 3D10 subclone is a fully differentiated eosinophilic myelocyte.
Core Specifications
  • Organism:Homo sapiens (Human).

  • Cell Type: Hematopoietic-Myeloid Leukemia; specifically an eosinophilic myelocyte to metamyelocyte.

  • Morphology: Round, non-adherent cells containing abundant eosinophil secondary granules.

  • Biosafety Level: BSL-2.

  • Doubling Time: Approximately 24–36 hours (some sources report up to 48 hours).

Culture Conditions
  • Growth Medium:RPMI 1640 supplemented with 8% Fetal Bovine Serum (FBS), 2mM L-glutamine, 1mM sodium pyruvate, and 50μM (or 5 x 10⁻⁵ M) Beta-mercaptoethanol.

  • Seeding Density: Maintain between 3 x 10⁵ and 1 x 10⁶ cells/mL.

  • Critical Threshold: Do not exceed 1 x 10⁶ cells/mL. Overgrowth causes degranulation, which releases toxic proteins (like MBP) that are fatal to the culture.

  • Subculturing: Split every 3–4 days (typically 1 volume old culture to 2 volumes fresh media).

Key Biological Markers
  • Granule Proteins: Expresses high levels of major basic protein (MBP), eosinophil peroxidase (EPO), and eosinophil-derived neurotoxin (EDN).

  • Receptors: Expresses GM-CSF and IL-5 receptors. Note that this subclone has reportedly lost the α-subunit of the IL-3 receptor.

  • Growth Support: Does not require exogenous cytokines for proliferation but responds rapidly to GM-CSF and IL-5.

Handling & Storage
  • Cryopreservation: 90% FBS + 10% DMSO.

  • Thawing Caution: Initial viability is often low (20%–30%) due to granule protein toxicity. Frequent medium replacement during the first 72 hours facilitates recovery.
