BioVector TM pGhost9:ISS1（有时也写作 pG+host9:ISS1）是一种用于革兰氏阳性菌（如乳酸菌、链球菌）基因失活和转座突变的温度敏感型穿梭载体。

• 选择标记：红霉素抗性 (ErmR)。在 E. coli 中的工作浓度通常为 150-200 μg/ml，在革兰氏阳性宿主中约为 5 μg/ml。

• 转座元件： 携带 ISS1（一种插入序列），能够通过复制性转座机制高效、随机地整合到宿主染色体中。

• 常用宿主：E. coliMC1061（克隆宿主），以及多种乳酸菌和链球菌。

2. 基因敲除/失活： 克隆目标基因的内部片段到载体中，通过升温强制质粒发生单交叉整合 (Single-crossover integration)，从而破坏目标基因。

• 转化与扩增： 在 30°C 下进行转化和筛选，以维持质粒的游离状态。

• 诱导整合： 将培养温度提升至 37°C 或 40°C，并保持红霉素压力。此时质粒无法自主复制，只有整合到染色体上的菌株才能存活。

• 效率参考： 在嗜热链球菌中，转座频率可达约 2 x 10⁻²。

• Replicon:repA (thermosensitive). It replicates at permissive temperatures (30°C) and is lost or forced to integrate at non-permissive temperatures (37°C–42°C).

• Selection Marker:Erythromycin resistance (ErmR); typical working concentration is 200 μg/ml for E. coli and 5 μg/ml for certain Gram-positive hosts.

• Host Strains: Primarily used in lactic acid bacteria; common cloning host is MC1061.

• Mutagenesis Mechanism: Utilizes the ISS1 (Insertion Sequence) element for random, replicative transposition into the host genome.

• Temperature Sensitivity: The plasmid contains a temperature-sensitive version of the pWV01 replication protein (RepA). At 37°C or higher, replication ceases, making it a "suicide" vector unless it integrates into the chromosome.

• Transposition Efficiency: High frequency of transposition (e.g., ~2 x 10⁻² in S. thermophilus) with random chromosomal distribution.
