主要菌株技术参数

不同菌株根据其遗传特性和代谢侧重点有所区别：

DJ 菌株 (NTCC BAA-1303)：最常用的实验室标准菌株，广泛用于基因功能研究。它是 OP 菌株的衍生物，转化效率极高，由于产生固氮菌素（Azotobactin），在平板上常呈现典型的绿色。
O 菌株：原始“野生型”菌株。其特点是菌落呈黏性或“胶状”，这源于其极高的海藻酸盐产量。
CA (或 OP) 菌株 (NTCC 13705)： O 菌株的“非黏性”变体，通常用于研究固氮酶系统的生化特性。
DSM 2289 (NTCC 478)：标准模式菌株，主要用于分类学鉴定和基础培养实验。

生长与培养条件

生物安全等级： BSL-1（非致病性，对人体安全）。
最佳生长温度： 26°C - 30°C。
培养基：常用 Burk’s 培养基（无氮培养基）。若需大量生长，可添加约 2.5% 的蔗糖或甘露醇作为碳源。
形态特征：细胞呈大卵圆形（2–4 μm），常成对出现。在逆境下可形成胞囊（Cysts），具有较强的抗干燥能力。
氧气耐受：专性需氧菌，但具有极高的呼吸速率，以消耗细胞内的氧气从而保护其对氧敏感的固氮酶。

Azotobacter vinelandii is a large, Gram-negative, aerobic bacterium primarily known for its ability to fix atmospheric nitrogen and produce valuable biopolymers like alginate and polyhydroxyalkanoates (PHB).

Strain Profiles

Different strains of A. vinelandii are used depending on whether the research focus is on genetics, biofertilizer production, or industrial bioplastics.

Strain DJ (NTCC BAA-1303): The standard laboratory strain for genetic studies. It is highly competent for taking up foreign DNA and is a derivative of the nongummy OP strain. It typically turns agar plates green due to the production of the siderophore azotobactin.
Strain O: The "wild-type" or original strain, characterized by viscous, gummy colonies due to high alginate production.
Strain CA (or OP) (NTCC 13705): A "nongummy" version of strain O that is frequently used for studying nitrogenase systems.
DSM 2289 (NTCC 478): A type strain used for taxonomic reference and general cultivation.

Biosafety Level: BSL-1 (Non-pathogenic).
Optimum Temperature: 26°C to 30°C.
Growth Media:Modified Burk's Medium is the standard for nitrogen-free growth. On solid media, it may require sucrose or mannitol (approx. 2.5%) as a carbon source.
Morphology: Large oval cells (2–4 μm), often appearing in pairs or clusters. It can form cysts (resting cells) resistant to desiccation.
Oxygen Tolerance: While it is an obligate aerobe, it has high respiration rates to "protect" its oxygen-sensitive nitrogenase enzymes.
