BioVector®MUSIi001-A-2 is a human induced pluripotent stem cell (iPSC) line genetically engineered to be hypoimmunogenic for use in allogeneic cell therapy research.

Key Characteristics
  • Genetic Modifications: It is a dual-gene knockout line created using CRISPR/Cas9 technology. It features biallelic disruptions of the B2M (Beta-2-Microglobulin) and CIITA (Class II Major Histocompatibility Complex Transactivator) genes.

  • Functionality: These modifications eliminate the expression of both HLA Class I and HLA Class II molecules on the cell surface. This makes the cells "invisible" to certain parts of the immune system, reducing the risk of graft rejection in allogeneic (off-the-shelf) therapies.

  • Parental Line: It was derived from the MUSIi001-A-1 line (a B2M-only knockout), which itself originated from the wild-type MUSIi001-A line. The original parental cells were fibroblasts from a 33-year-old healthy female.

  • Pluripotency: The line maintains typical iPSC characteristics, including the ability to differentiate into all three embryonic germ layers.
