The U-CH2 cell line is a human adherent cell line derived from a recurrent sacral chordoma, a rare type of slow-growing bone tumor. It is a vital research model characterized by specific morphological and molecular features typical of chordoma. U-CH2 Cell Line Datasheet Organism: Homo sapiens (Human) Tissue: Sacrum (bone/mesenchymal tissue) Disease: Chordoma (recurrent sacral) Sex: Female Age: 72 years at time of isolation Morphology: Heterogeneous, consisting of physaliferous cells (vacuolated cells with mucinous substance), which are typical chordoma features. Growth Properties: Adherent monolayer, grows relatively slowly. Doubling Time: Approximately 3-7 days. Key Marker: Contains amplification and positive expression of the transcription factor Brachyury (T), the most specific marker for chordoma. Biosafety Level: Typically BSL-1. Culture Conditions Culture Medium: A 4:1 mixture of Iscove's Modified Dulbecco's Medium (IMDM) and RPMI-1640 medium, supplemented with 10% Fetal Bovine Serum (FBS). Culture Flasks: The cells should be cultured in coated flasks (e.g., coated with 0.1% gelatin) to facilitate attachment. Growth Temperature: 37°C. CO₂ Concentration: 5% CO₂ in a humidified incubator. Subculture Routine: Rinse the cell layer with calcium and magnesium-free Dulbecco's phosphate-buffered saline (D-PBS). Use Trypsin-EDTA solution for detachment (usually within 5 to 15 minutes). Cells may need to be placed at 37°C to facilitate dispersal. Avoid agitating the cells excessively to prevent clumping. Passage when approximately 80-90% confluent, typically a 1:3 to 1:5 ratio is recommended. Storage and Handling Storage: Cryopreserved vials should be stored in the vapor phase of liquid nitrogen, not at -70°C, to ensure long-term viability. Thawing: Thaw the vial rapidly in a 37°C water bath, then immediately transfer the contents to a centrifuge tube containing fresh, pre-warmed complete culture medium to remove the cryopreservation agent and avoid excessive alkalinity.
