The NTCC™ OKF6/TERT-2 cell line is a human cell line of oral epithelial keratinocytes that has been immortalized through the introduction of the hTERT (human telomerase reverse transcriptase) gene. This makes it a valuable and reproducible model for studying the normal oral mucosa.

Key Characteristics
Origin: The parent line, OKF6, was derived from the floor of the mouth of a 57-year-old male. The TERT-2 designation indicates a specific immortalized sub-line.
Immortalization: Unlike primary cells, which have a limited lifespan, the OKF6/TERT-2 cell line expresses telomerase, which allows it to proliferate indefinitely without becoming cancerous. This provides a stable and consistent source of cells for long-term studies.
Genetic Profile: A key feature is a homozygous deletion of the p16INK4a and p14ARF genes. These are well-known tumor suppressor genes, and their absence contributes to the cell line's extended lifespan.
Physiological Relevance: The cells are able to differentiate in vitro into a multilayered epithelial structure that closely resembles the normal human oral mucosa, complete with a basal layer, stratum spinosum, and a non-keratinizing stratum corneum. This ability to form a tissue-like structure makes it a more physiologically relevant model than many other cell lines.
Research Applications
The OKF6/TERT-2 cell line is an important tool in various fields, particularly for studies that require a reliable model of the oral cavity. Its main applications include:
Toxicology & Safety Testing: It is used to evaluate the irritant, inflammatory, or damaging effects of consumer products like toothpastes, mouthwashes, and e-liquid aerosols on the oral mucosa.
Oral Biology & Disease Modeling: Researchers use it to study the molecular biology of diseases affecting the oral cavity, such as periodontal disease and oropharyngeal malignancy. It is also used for wound-healing assays to understand tissue repair mechanisms.
Drug & Gene Delivery: Its ability to form a functional epithelial barrier makes it useful for testing the delivery of drugs or genes through the oral mucosa.
3D Organotypic Models: The cell line is a core component of advanced 3D organotypic models, where it is cultured with fibroblasts to create a full-thickness tissue analog that mimics the native oral mucosa. This allows for the study of complex cell-cell interactions.
