PhCMV-driven expression of hTRL1068 heavy and light chains linked by a F2A self-processing peptide and a furin cleavage site (RKRR) (PhCMV-hTRL1068-HC-RKRRF2A-LC-pA). hTRL1068-HC-RKRRF2A-LC was generated by annealing PCR fragment of TRL1068-HC using OYL170 (5'-CCCCCCAAGCTTTCGCCCACCATGGACTGGACCTGGAG-3', HindIII underlined) and OYL201(5'-AGGTCCAGGGTTGGACTCCACGTCTCCCGCCAACTTGAGAAGGTCAAAATTCAAAGTCTGTTTCACGGGGGCCCGTCTCTTCCGTTTGCCGGGGCTCAGGG-3', RKRRF2A sequence underlined) with PCR fragment of TRL1068-LC using OYL203 (5'-ACCTTCTCAAGTTGGCGGGAGACGTGGAGTCCAACCCTGGACCTGAAGCCCCAGCGCAGC-3') and OYL173 (5'-CTAGTCTAGACTACTAGCACTCGCCCCG-3', XbaI underlined). The annealing product was then digested and cloned into pcDNA3.1(+) (HindIII/XbaI).
