pCzn1 Plasmid + Arctic Express Host Cold Expression SystemExperiment description The target gene was obtained by gene synthesis, cDNA cloning, and reverse transcription, and the target gene was subcloned into the expression vector pCzn1. The target gene was fused with His-tag protein at the N-terminus, and the subclone was confirmed to be correct by sequencing and enzyme digestion. Expression plasmids. Using the constructed expression plasmid, the Arctic Express host strain was transformed. Under 11 degree cold IPTG induction conditions, the target protein expression was detected by SDS-PAGE to confirm that the protein was distributed in the supernatant or in the precipitate. If the target protein product is distributed in the supernatant, the target protein is obtained by affinity purification on the Ni column; if the target protein is distributed in the precipitate, the target protein is re-dissolved after elution through the gradient of the inclusion body, and if necessary, the resolubilized protein is obtained. The purity is further increased by affinity purification.
