around 30 different transient vector configurations, which we identified by testing over 250 different combinations of promoters, introns, polyadenylation sequences and viral amplifiers in HEK and CHO cell lines.The reason we have so many vectors is that even similar cell lines seem to have different control element preferences, as is shown in the graphs below. Added to this, differences in desired duration of culture and the localization of the protein being produced (intracellular, membranebound or secreted) make it impossible for a single vector to meet all needs.
