The bacteriophage P1 cloning system was originally developed as an alternative to YAC
and cosmid systems for cloning high-molecular-weight genomic DNA. An in vitro phage
packaging system is used to generate >105
clones containing inserts up to ∼95 kb in size
from size-fractionated insert DNA. Introduction of recombinant DNA into the cell in this
manner distinguishes this method from more recently developed systems that rely upon
bacterial electroporation to introduce large-insert DNA.
