pTX4577 is a suicide plasmid used in the construction of isogenic hyl-deficient mutants in Enterococcus faecium. It has been used in studies to investigate the virulence of Enterococcus faecalis in mice and rabbits:
- The pTX4577 plasmid was used to construct an isogenic hyl-deficient mutant strain.
- The mutant strain and the wild type strain TX2466 were cultured to compare their growth curves.
- Both the mutant and wild type strains were cultured on solid media with kanamycin to test their tolerance to the antibiotic.
- The virulence of the strains was studied in models of mice peritonitis and rabbit endocarditis.
- The mutant strain was identified by PCR, pulsed field gel electrophoresis, and Southern blotting.
The hyl gene is considered a major virulence factor in Enterococcus faecalis and is important in the pathogenesis of the bacterium.
