pAdenoVator-CMV5-IRES-GFP is a new Ad transfer vector that combines the sgGFP (SupergloTM Green Fluorescent Protein) with the AdenoVatorTM technology. In this vector the GFP is co-expressed with the gene of interest in an IRES (Internal Ribosome Entry Segment) expression cassette (Mosser et al., 1997). The IRES is a sequence that supports translation initiation from the second cistron in a dicistronic message. In this transfer vector both the gene of interest and the GFP are placed under the control of the modified CMV5 promoter, for very high expression. This arrangement allows for easy visualization of the transduced cells and maximal expression levels with lower MOIs (multiplicity of infection). In addition, inclusion of the GFP in this new transfer vector offers a way to accelerate the identification of Ad recombinants and to facilitate the functional characterization of genes both in vitro and in vivo by allowing easy quantification of gene transfer and expression in a non-invasive manner. Please note that expression levels of the two genes is not equimolar; from our own experience we estimate the proportion of genes expressed to be 3:1 (1 being the gene following the IRES i.e. the GFP in our construct).
