pNBU2-bla-ermGb chromosomal insertion vector (Structure 16: 1-11). Derivative of pKNOCK-bla-ermGb into which a PCR-amplifiedintN2-att fragment (J. Bacteriol. 182: 3559-71) was inserted using engineered SalI and KpnI ends. MCS sites can be used to insert fragmentsinto pKNOCK-Nbu2-ermb and the entire plasmid construct can then be conjugated into Bacteroides spp. and insertions made into either of thetwo, NBU2-targeted, tRNAser loci. Uses include complementation of IDM, deletion or transposon mutants and introduction of exogenous DNAfragments into the genomes of Bacteroides spp. Select restriction sites are shown along with the multiple cloning site (MCS), R6K replicationorigin (oriR6K) and the RP4 transfer origin (RP4 oriT).
