Ad5/F35MaxTM vector packaging system based
on AdMax packaging system. Ad5/F35 is a chimeric adenovirus vector for the Ad5 virus fiber was modified into a type of the
35 adenovirus. In the genome only fiber gene is chimeric, and other genes still retain the characteristics of Ad5 vector. The
packaging system includes pBHG-fiber5/35 scaffold plasmid, shuttle plasmid and HEK293 cell. The mechanism is similar to
the AdMax system. After transfected into HEK-293 cells, the pBHG-fiber5/35 scaffold plasmid can match to the shuttle
plasmid carrying loxP site, and incur site directed recombination by Cre-loxP system, which can produce recombinant
adenovirus (Ad5/F35) with exogenous genes. The recombinant adenovirus obtained by this way is E1 defect replication
defective adenovirus, which can only express exogenous gene, but not be able to amplify itself in cell without E1 region. The
characteristic of Ad5/F35MaxTM adenovirus packaging system is to complete recombinant virus maturation by recombinant
enzyme in eukaryotic cell, which not only retains the advantages of quick virus maturation, good stability and high yield of
AdMax system, but also has advantages of wider infected cell range, higher infection efficiency (higher infection efficiency in
various hematopoietic cells and tumor cells in Ad5/F35 than that in common Ad5), more exogenous gene expression, and
efficient and stable packaging system.
