Plasmid pSC11 was generated by inserting an XbaI-SmaI fragment of pSC10 containing the lacZ gene under the control of the P11 promoter into the HincIl site of pGS53, which contains the promoter of the 7.5K polypeptide gene.
The important features of pSC11 are the presence of the lacZ gene under the control of the 11K promoter, a second vaccinia virus promoter derived from a vaccinia virus gene that encodes a 7.5K polypeptide, a unique SmaI site downstream of the 7.5K promoter for the insertion of foreign protein-coding sequences, and
flanking vaccinia virus TK sequences for homologous recombination. The two promoters were oriented in opposite directions to prevent the possibility of polarity effects.
