Raji cells were used in most cell-binding experiments. Raji is a Burkitt lymphoma cell that expresses MHC classes I and II and is a very effective presenter of enterotoxins . RM3 is a derivative of Raji that has lost class II expression.
The RM3 cell line was utilized for HLA transfections. RM3 is derived from the human B lymphocyte cell line Raji (DRA1*01:01, DRB1*03:01, DRB1*10:01, DRB3*02, DQA1*01:01, DQA1*05:01, DQB1*02:01, DQB1*05:01, DPB1*01:01) following mutagenesis with ethane methylsulfonate and selection for class II negative cells. Transfection of RM3 cells to produce cell lines expressing a single MHC specificity has been utilized in previous studies.

RPMI 1640 medium supplemented with 2 mM glutamine, 1% (vol/vol) nonessential amino acids, 1% (vol/vol) sodium pyruvate, penicillin (50 U/ml), streptomycin (50 μg/ml) (all from Life Technologies) and 10% heat-inactivated fetal bovine serum.
