The plasmid (pAAV-HBV1.2) for hydrodynamicinjection contains a 1.2 over length HBV genome spanning nucleotides 1,400–3,182/1–1,987 flanked by inverted terminal repeats of an adeno-associated virus. The plasmid contains the coding sequence for HBV genotype A, serotype adw. The 1.2-full-length HBV genome flanked by AAV2 ITRs was packaged into capsids from AAV8 after cotransfection of HEK 293 cells with pAAV-HBV.
