Selection and characterization of phleomycinresistant
colonies after direct transformation of Candida guilliermondii
with plasmid pG-Sable. Structure of pG-Sable (* indicates single-site
restriction enzymes) and selection of C. guilliermondii cells (ATCC
6260) on phleomycin-containing plates after transformation with
either sterile water (control) or 10 lg of linearized pG-Sable. Plates
were incubated for three days at 35 °C. The ApaI unique site was
used for linearization before transformation. The gel image shows the
results of yeast colony PCR on a subset of 16 randomly selected
clones performed using specific primers (BleS and BleR) to amplify the
Sable sequence.
