SSA-RPG reporter双报告基因质粒，组成型表达DsRed红色荧光，当被打断 的puro被核酸酶修复后条件性表达puro筛选基因和EGFP绿色荧光报告基因，用于检测转染效率和富集遗传修饰细胞。

Constitutively express DsRed and conditionally express Puro and EGFP. Used to test transfection efficiency and enrich genetically modified cells.
Puromycin and EGFP will not be expressed until the interrupted puro gene is repaired by single strand annealing (SSA) pathway after a nuclease generates double strand breaks (DSBs).
Nuclease target sequence can be annealed and inserted into the backbone between BamHI and NotI sites using restriction enzymes or Gibson cloning methods.
